Can this be done on Mythic+? For example a Mythic with a Level 2 Key?

Asking because some of these quests only give credit on Normal Mythic, for example one of the Eye of Azhara Mythic quests only gives credit on the baseline mythic.

If you deleted your "Black Rook Missive" you will not be able to turn in the quest as it is required. If you put in a ticket and state that you were not aware that the item was required and you lost it they may restore it but they might have you drop the quest and do it again. It's important to hold onto these items. Good luck.

If after you complete the fungeon it says you still need the missive, you can go and turn it in. I was confused and even made a ticket. I went back to my order hall and saw the yellow ? on my mini map, but when I went to turn it in, the quest was greyed out like I still did not complete it. I clicked it anyway and was able to turn it in and get my box.
