These drop off virtually any roaming beast found in Shadowmoon Valley (but not on Netherwing Ledge) -- feboars, dimetradons, chimaeras, scorpions, etc.
When placing the poison, be sure to do so one at a time, wait for quest credit, then do the next. Trying to do multiple poisons at once, seems to only grant credit for one.
I also recommend you farm one extra poison and save yourself a trip back to the mainland. Chances are fairly high you won't get credit for one of the poisons you drop and you'll find yourself at 11/12. At a minimum, you can just use the extra gland the following day.

Better get 13 Fel Glands, to be sure. I put one poisoned mutton near orcs, but they just do nothing, so i don't get credit for that one.

To clear things up:
1) You can't get more than 12 of these. They are quest items and capped at 12. No more will drop when you have 12 in your inventory.
2) To avoid wasting them, never drop any next to a peon camp where the peons are standing completely still. These peons are evade bugged and will not give you quest credit. Only drop them where peons are actually mining the crystals.

(1) Want to stock up for the next day? Get 12, do the quest, but DO NOT turn it in yet. Go out, kill more mobs, get 12 more, then go turn it in. Your extra fel glands will not disappear when you turn in the quest.
Aparently, you can't get any more fel glands after you turn in the quest. I tried to get them from the same mobs i got the first few before I turned in the quest just to test it out.
(2) you don't have to be right next to the peons, just as long as you're pretty close. You can also be running and click on the Mutton and it will place it on the ground. I just run by, place it without stopping while running towards a new group.

I find that the best place to farm these is in the small felfire pool outside Eclipse Point, the diemetradons there respawn quickly and are easy to kill.

As the Fel Glands are a quest item you can only collect them while you are on the quest.

The best drop rate is ~20%, so you'll need to kill around 60 mobs to obtain all 12.
Yeah, best to do this with 2 or 3 others.

Best place I've found so far is west of the aldor base, about a 90% drop rate off the felboars.

Generally the best drop rate for Fel glands is from Netherskates

not Fel boars or Greater Felfire Diemetradon since I tested it many times with 4 chars. If u look at percentage at wowhead drop rate you can notice that boars and diemetradons have 21,18,and17% and for Netherskates is 15% but amount of killed mobs is 3-4 times larger because many people kill without quest since they are in easy accesable areas and the Netherskates are mostly on high grounds and are reached with flying mount.
One more plus for Netherskates is that they also drop motes of mana and next to them are Air elementals who drop motes of air.So, less then 10 minutes is needed to get 12 glandes and maybe couple of primals. My favorite place is just south of Dragonmaw fortress, they are easy to kill and not many people go there.

As a paladin, I've found Felboars to be the way to go. Just west from the Altar of Sha'tar, and just east from the Warden's Cage, between the Cage tne the Black Temple, there are two groups that may not respawn fast enough, but it's good if you clear one, and go to the other. Exorcism adds a huge boost to my overall damage, and I prefer Felboars instead of the lizards or the chimeras.

Go to the Fetid Pool !
where is it? : Hit your map key, see the green pool of water on the eastern side of Legion Hold ? thats it.
whats there? : an underwater quest objective you need to loot from an earlier quest ( the remains of a fel reaver ) and lots of Mutant Horrors.
whats so good about it? : well, the mutants here are level 67, so they are green at 70, thus, they die fast. They respawn faster than you can say, "christ, these respawn fairly quick!", and because of the mass numbers of them, they can be rounded up and AoE'd down in big groups, any half decent fury warrior can easily pull 5 or 6 of these guys, and spam your cleaves and whirlwinds and sweeping strikes.
The area is incredibly small, so you can literally round up -all- the mutants in one go there and wipe them out, they'll have fully respawned in about 5 minutes, tops, and thats an extremely long wait for them, I ran around in circles around the pool and never ran out of groups to kill.
And to top it off, they cast
mutated blood
on you in the fight, which deals relatively insignificant damage to you, while also allowing you to pump out 10% more damage over all :P Combine that 10% boost with all the AoE spammage that should be going on, and you'll rip through them in the blink of an eye, and have your 12 fel glands in no time.
The individual mob itself may have a lower droprate, but due to the small area, as mentioned, they can be AoE'd down so fast in groups ( even faster if you get mutated blood ) , that it will seem like you got the glands in no time at all.
i'm never going back to the felboar/dimetradon combo :)

wow i have the quest ive been killing these things 5 at a time and no fel glands, easy to kill sure enough sucky drop rate however

What I want to know is, why people keep talking about "stocking up for the next day". It...really doesn't make a difference. You're saying you farm up the 12, use the poisons, then farm 12 more for tomorrow. Tomorrow you grab the quest, poison, then farm 12 for the next day? How is it any different from just farming after you grab the quest each day?
I guess if you know you won't have time to farm the next day, that's fine, but if you're farming again for the following day, there's not much point, except maybe to give you a placebo effect of working ahead.

Things must have changed with patch 3.0.8. I got a 100% drop rate for these off the dimetradons around the pool this morning. 12 kills - 12 glands!

Yes as of patch 3.0.8 droprate of Fel Gland has been increased to 100%. Also the droprate of Netherwing Relic has been increased to 100% as well.

Closest, well populated mobs for Fel Glands 46, 61 Eclipse Point. Plenty of Felfire Diemetradons.

Stocking up for the next day does not work anymore, not for me nor the user @Jarynelle who first posted this:
"For anyone doing this post-cata, stocking up doesn't seem to work anymore. I couldn't get more fel glands to save for the next day. (Quest was complete, but not turned in - as stated above)"
